A near infrared fluorescent polymeric drug delivery system (NIRF DDS) with pH-responsive drug release properties has been designed and developed. This material was prepared by chemical conjugation of the anticancer drug doxorubicin and hydrophobic aminocyanine dye to triblock copolypeptide via hydrazone and amide bonds, respectively. Conjugation with aminocyanine shows almost no toxicity of the material, while conjugation with doxorubicin induces pronounced toxicity on the original biocompatible material. The pH sensitive drug release nature of the near infrared fluorescent polymeric drug (NIRF prodrug) was confirmed by accelerated drug release at pH of 5.0 via an in vitro drug release experiment and gradual drug cleavage from the NIRF prodrug during a confocal laser scanning microscopic (CLSM) experiment. The CLSM experiment also reveals that the released drug subsequently migrated to the nucleus, while the polymeric residue still remained in cytoplasm, indicating that the as-prepared polymer can be a promising candidate for theranosis of cancer.

To address this problem, we designed and developed Schiff-base linked imidazole modified polylysine with a reversible-PEGylation catiomer (SL-ImPEG-SS-PLL) for high efficiency gene delivery. The reversible PEGylation was introduced for in vivo circulation, as well as selective PEG detachment to augment the cellular internalization, while introducing Schiff-base linked imidazole residues into polylysine was expected to accelerate the endosomal escape of the DNA payload, as well as facilitate intracellular DNA unpacking and release, thus significantly enhancing gene delivery efficiency. The size alteration of the SL-I15mPEG-SS-PLL/pDNA polyplexes in the presence of 10 mM GSH suggests stimulus-induced PEG detachment under tumor relevant reduction conditions. Acid-base titration assays indicate that imidazole residues confer on polylysine remarkable buffering ability. More importantly, in vivo transfection of pEGFP reveals that GFP expression was found not only in some of the important organs, such as the liver, spleen, kidneys and lungs, but also in the transplanted carcinoma. These experimental results suggest that the reversible PEGylation and Schiff-base linked imidazole modification make SL-ImPEG-SS-PLL a great potential candidate for an effective and biocompatible gene delivery system. Two types of novel Polyhedral Oligomeric Silsesquioxanes respectively containing hydroxyl group and epoxy group (P-POSS and E-POSS) were achieved and evaluated. The structure had been characterized by IR spectra and NMR spectra. Dicyclopentadiene bisphenol dicyanate ester (DCPDCE) composites were then prepared using P-POSS and E-POSS respectively. Their effect on the curing kinetics, dielectric, mechanical, flame-retardant and thermal properties and water absorption of the resulting composites were investigated. The results suggested that the addition of modified POSS could facilitate the curing reaction of DCPDCE. Besides, the DCPDCE composites containing modified POSS exhibited excellent flame-retardant property over pure DCPDCE resin. Adding only a little amount as small as 1.5 wt% P-POSS or 2.5 wt% E-POSS could change the UL-94V of DCPDCE resin from V-2 to V-0. The composite with P-POSS exhibited better flame-retardant and thermal properties than the composite with E-POSS. However, composite filled with E-POSS presented better dielectric property and lower water absorption.
